This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human CD30 / TNFRSF8 and conjugated with FITC under optimum conditions, the unreacted FITC was removed.

背景

简介
Mouse anti Human CD30, clone C03-8D3, recognizes the human CD30 cell surface antigen also known as TNFRSF8. CD30 is a 120 kDa, type I transmembrane glycoprotein of the tumour necrosis factor (TNF) receptor superfamily. CD30 was originally identified as a molecule expressed on Hodgkin and Reed Sternberg cells in Hodgkin’s disease but has subsequently been detected on certain non-Hodgkin's malignancies as well as some virally transformed cells. It is also expressed on a subset of activated human T-cells, and by a few extrafollicular T and B-cell blasts in normal lymph nodes.

FITC标记的CD30抗体是一类可以特异性结合CD30的多克隆抗体,主要用于检测CD30的Western Blot、IHC-P、IF、ELISA、Co-IP等多种免疫学实验。
检测原理
双抗体夹心法测定标本中CD30水平。用纯化的CD30抗体包被微孔板,制成固相抗体,往包被单抗的微孔中依次加入CD30,再与HRP标记的CD30抗体结合,形成抗体-抗原-酶标抗体复合物,经过彻底洗涤后加底物TMB显色。TMB在HRP酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的CD30呈正相关。用酶标仪在450nm波长下测定吸光度(OD值),通过标准曲线计算样品中CD30浓度。
