Activity
Measured by its binding ability in a functional ELISA. When Recombinant Human Cerebellin-2 is coated at 5 μg/mL, biotinylated recombinant rat NRXN-1 beta binds with an apparent K D <1.5 nM. Measured by its ability to enhance neurite outgrowth of E16-E18 rat embryonic cortical neurons. Recombinant Human Cerebellin-2, immobilized at 3-30 μg/mL, is able to significantly enhance neurite outgrowth.

Background: Cerebellin-2
Cerebellin-2 (CBLN2) is an approximately 25 kDa N-glycosylated protein in the Cerebellin family of TNF superfamily molecules (1). Cerebellins contain a collagenous domain and a C-terminal TNF/C1q-like domain. Human Cerebellin-2 consists of this 175 amino acid (aa) region following an N-terminal 28 aa potential cytoplasmic domain and a 21 aa potential transmembrane segment. Within the Cerebellin-like region, human Cerebellin-2 shares 99% aa sequence identity with mouse, rat, and chick Cerebellin-2 (2‑4). It is expressed in multiple regions of the nervous system including the neocortex, olfactory bulb, thalamus, hypothalamus, midbrain (5), preganglionic sensory nerves, dorsal root ganglia, and sensory and motoneurons of the spinal cord (4, 6) as well as in the zona glomerulosa of the adrenal cortex (7). Cerebellin-2 forms cell associated homo-oligomers and heteromers with Cerebellins-1, -3, and -4 [Iijima 1049]. It binds with high affinity to alpha and beta Neurexin 1 isoforms that contain the splice site 4 insert (+SS4) and more weakly to Neurexin 2 beta (+SS4) and Neurexin 3 beta (+SS4) (9, 10). These interactions promote presynaptic differentiation in cultures of cortical and cerebellar neurons (9, 10).
