General description

磷酸钙法转染是将DNA引入真核细胞的常用方法。这一技术已经在广泛的细胞类型中使用，可实现瞬时和稳定的转染效果。

The protocol was developed for transient transfection of CHO cells using pSV40-CAT plasmid as a reporter gene.

Principle

该操作方案要求缓慢混合含有磷酸钠的HEPES缓冲盐及含有DNA的CaCl₂溶液。之后会形成DNA-磷酸钙共沉淀，这些沉淀附着在细胞表面并被细胞吸收（大多数通过内吞作用吸收）。
